Diaz has been out injured since October and suffered a setback in his recovery, but was back in first-team training this week.
There was some hope he might feature at the Etihad Stadium, but Klopp confirmed the game has come too soon for the ex-Porto man.
Klopp: Luis back in small parts of team training after a long time out," Klopp said of Diaz. "[It's] fantastic to have him back but he won't train with us today. This week was a first step. Next week I think we will increase it and then he's ready.
"Darwin couldn't train for two or three days this week, but trained yesterday. He is back in contention," the manager added.
